In allogenic and xenogenic transplantation, adequate immunosuppression plays a major role in graft survival, especially over the long term. The effect of immunosuppressive drugs on neural stem/progenitor cell fate has not been sufficiently explored. The focus of this study is to systematically investigate the effects of the following four different immunotherapeutic strategies on human neural progenitor cell survival/death, proliferation, metabolic activity, differentiation and migration in vitro: (1) cyclosporine A (CsA), a calcineurin inhibitor; (2) everolimus (RAD001), an mTOR-inhibitor; (3) mycophenolic acid (MPA, mycophenolate), an inhibitor of inosine monophosphate dehydrogenase and (4) prednisolone, a steroid. At the minimum effective concentration (MEC), we found a prominent decrease in hNPCs' proliferative capacity (BrdU incorporation), especially for CsA and MPA, and an alteration of the NAD(P)H-dependent metabolic activity. Cell death rate, neurogenesis, gliogenesis and cell migration remained mostly unaffected under these conditions for all four immunosuppressants, except for apoptotic cell death, which was significantly increased by MPA treatment.  相似文献

Telomerase is a ribonucleoprotein with an intrinsic telomerase RNA (TER) component. Within yeasts, TER is remarkably large and presents little similarity in secondary structure to vertebrate or ciliate TERs. To better understand the evolution of fungal telomerase, we identified 74 TERs from Pezizomycotina and Taphrinomycotina subphyla, sister clades to budding yeasts. We initially identified TER from Neurospora crassa using a novel deep-sequencing–based approach, and homologous TER sequences from available fungal genome databases by computational searches. Remarkably, TERs from these non-yeast fungi have many attributes in common with vertebrate TERs. Comparative phylogenetic analysis of highly conserved regions within Pezizomycotina TERs revealed two core domains nearly identical in secondary structure to the pseudoknot and CR4/5 within vertebrate TERs. We then analyzed N. crassa and Schizosaccharomyces pombe telomerase reconstituted in vitro, and showed that the two RNA core domains in both systems can reconstitute activity in trans as two separate RNA fragments. Furthermore, the primer-extension pulse-chase analysis affirmed that the reconstituted N. crassa telomerase synthesizes TTAGGG repeats with high processivity, a common attribute of vertebrate telomerase. Overall, this study reveals the common ancestral cores of vertebrate and fungal TERs, and provides insights into the molecular evolution of fungal TER structure and function.  相似文献

Non‐consumptive predator effects may have dramatic consequences for host–parasite interactions by influencing the ability of prey items to avoid, resist, or tolerate infection. Both predators and parasites can affect host traits, such as growth rates and behavior, and these effects may in part be mediated through shared physiological pathways (e.g. the glucocorticoid stress hormone, corticosterone [CORT]). Here, we examined the effects of trematode parasites (Digena: Echinostomatidae) and predator (larval odonate) exposure on larvae of two amphibian species (Rana sylvatica and R. clamitans) in laboratory experiments. First, we measured behavior and CORT responses of tadpoles exposed to predator chemical cue in combination with parasite cue or under direct exposure to parasites. We then measured the combined effects of predator cue and parasite infection on survival and traits. Evidence for effects of parasite cue in our study was equivocal, but we found novel interactive effects of parasites and predators on larval frogs. Parasites and predators had antagonistic effects on CORT, behavior, and morphology, and negative synergistic effects on development. In addition, parasite infection and predator cues additively reduced activity levels of both species and growth in wood frogs. Negative effects of parasite infection on survival and traits were dose‐dependent for both species, although wood frogs generally experienced stronger effects of infection than green frogs. Our results emphasize the importance of considering effects of parasites as well as predators, since both can have strong effects on survival and the combination can have both additive and non‐additive effects on key traits. These effects likely have important implications for amphibian population dynamics, community structure, and conservation.  相似文献

The enigmatic fissure deposits of south‐western England and southern Wales are famous for their unique assemblage of Late Triassic vertebrates, although their age is contentious. While recent studies of palynomorphs have dated some as Rhaetian, their conchostracan (Crustacea, Branchiopoda) assemblages have not been described in detail nor used in biostratigraphy. We find that species determination of British Late Triassic conchostracans requires detailed observations of size, shape and ornamentation. We provide evidence that although Euestheria brodieana is invariably smaller than E. minuta, with some slight differences in carapace ornamentation, the traditional view that they are very similar is upheld. The use of conchostracans as a biostratigraphical tool is here tested by application to the British Triassic fissures at Cromhall quarry where the usual stratigraphical evidence provided by superposition is absent. We find no distinction between conchostracans from bedded Rhaetian deposits of the UK and specimens collected from the fissure deposits of Cromhall Quarry, Gloucestershire, supporting a late Rhaetian age for these deposits.  相似文献
